Procaine is a chemical. People use it for medicine. Be careful not to confuse the procaine used by mouth with the prescription procaine given by injection only under the supervision of a healthcare professional.

Procaine is most often used as an anti-aging agent for conditions such as dementia, age-related decline in memory and thinking skills, quality of life, and many others, but there is no good scientific evidence to support these uses.

As a prescription-only injection, procaine is used for local anesthesia.

Mechanism of action

Procaine acts mainly by inhibiting sodium influx through voltage gated sodium channels in the neuronal cell membrane of peripheral nerves. When the influx of sodium is interrupted, an action potential cannot arise and signal conduction is thus inhibited. The receptor site is thought to be located at the cytoplasmic (inner) portion of the sodium channel. Procaine has also been shown to bind or antagonize the function of N-methyl-D-aspartate (NMDA) receptors as well as nicotinic acetylcholine receptors and the serotonin receptor-ion channel complex.

a neuromuscular disease such as myasthenia gravis; or

Biochem/physiol Actions

Procaine is a Na⁺ channel blocker, commonly used as an anesthetic agent and is considered safer than cocaine. It is also useful as a painkiller to treat pain, associated with joints and tendons.

Procaine is well-known as a safe and nontoxic local anesthetic; however, its therapeutic value extends far beyond its local anesthetic effects. Chemically, procaine is an alcohol ester of p-amino benzoic acid (PABA), and is broken down and detoxified by the enzyme plasma cholinesterase, which occurs in all body tissues. It is metabolized so thoroughly that only a small part of it needs to be detoxified by the liver and only 2% is eliminated through the kidneys. In addition to PABA, (part of the vitamin B complex), procaine is broken down into diethyl-amino-ethanol. Both have antihistamine effects. PABA is an active agent against pathological sclerosing and hardening of tissues. Diethyl-amino-ethanol is a vasodilator, which improves blood flow and can lower blood pressure. It also has a mild stimulant effect on the central nervous system.
